Description

Paracetamol is N-acetyl para-amino phenol derivatives. It has analgesic, antipyretic and weak anti-inflammatory activity. Chemical formula is C8H9NO2 and its molecular weight is 151.2. Chlorpheniramine Maleate is a H1 -receptor antagonist, antihistamine. Chemically, it is (3RS)-3-(4-Chlorophen...

Clinical Pharmacology

Paracetamol, also known as acetaminophen, is an inhibitor of the enzyme cyclo-oxygenase and inhibits the synthesis of prostaglandins in the central nervous system and peripherally blocks pain impulse generation; produces antipyresis from inhibition of hypothalamic heat-regulating center. There...

Pharmacokinetics

Paracetamol is readily absorbed from the GI tract. Peak plasma concentration occurs at 10-60 minutes after oral dose. It is distributed into most body tissues. It crosses the placenta and present in breast milk. Plasma protein binding is negligible at usual therapeutic concentration but increase wit...

Chlorpheniramine is absorbed selectively and slowly from the gastrointestinal tract, peak plasma concentrations occurring about 2.5 to 6 hours after administration by mouth. It appears to undergo first pass metabolism, about 70%is bound to plasma proteins. It is metabolized primarily hepatic vi...

Phenylephrine Hydrochloride is readily and completely absorbed from the gastrointestinal tract. Following oral administration, peak plasma concentrations are achieved in 1-2 hours. The mean plasma half-life is in the range 2-3 hours. The volume of distribution is large (200 to 500 litres). Penetrati...

Contraindication

It is contraindicated in patients who have hypersensitivity to paracetamol, chlorpheniramine and phenylephrine; patients with severe hypertension or severe coronary artery disease or those receiving monoamine oxidase inhibitors (MAOI); and newborn and premature infants.

Precaution

Anti-histamine can cause reduced m...

Adverse Effects

The most common adverse effects are increased blood pressure, increased heart rate, dizziness, drowsiness, nausea, vomiting, dry mouth, loss of appetite, indigestion, abdominal pain, diarrhea, urinary retention, palpitation, tremor, muscular weakness and ringing sensation in ears.

Drug Interaction

MAOI : Concurrent administration may result in unpredictable increase in blood pressure by increased receptor activity.

Antihypertensives: Sympathomimetics may reduce the effect of antihypertensive drugs.

Alcohol: Increased risk of liver damage.

Storage

Store below 30ºC in cool, dry place. Protect from light and moisture. Keep out of reach and sight of children.
