First-line treatment of hypertension and myocardial ischaemia associated with stable angina pectoris. Used alone or in combination with other anti-hypertensive agents.
Adults:
Hypertension:
Initial dose: 2.5 - 5 mg once a day Maximum dose: 10 mg per day. Angina:
Initial dose: 5 mg once a day Maximum dose: l O mg per day.
Children (6 - 17 years):
Hypertension:
Initial dose: 2.5 mg once a day Maximum dose: 5 mg per day.
Children under 6 years must not be taken unless recommended by physician.
Elderly:
Initial dose: 2.5 mg once a day Maintenance dose: 2.5 - 10 mg once a day Maximum dose: 10 mg per day.
Dosing of elderly should start at the lower end of dosing range due to possible increased incidence of hepatic, renal or cardiac impairment. Elderly patients also show decreased clearance of amlodipine.
Each film-coated tablet contains:
Amlodipine Besilate B.P equivalent to Amlodipine ... 2.5 mg/ 5 mg/ 10 mg.
Amlodipine besilate is 3-Ethyl 5-methyl (4RS)-2- [(2-aminoethoxy)methyl]-4-(2-chlorophenyl)-6-methyl-1, 4-dihydropyridine-3,5-dicarboxylate benzenesulfonate.
It has an empirical formula ofC26H31ClN208S and molecular weight is 567.1.
Amlodipine is a dihydropyridine class of calcium channel blocker with a long duration of action, used for the treatment of hypertension and angina pectoris. It inhibits the passage of calcium through the voltage dependent L-(for 'long-opening') class membrane channels in cardiac muscle and conduct...
Amlodipine in general is well absorbed from the gastrointestinal tract and their systemic bioavailability depends on the extent of first-pass metabolism in the gut wall and liver. Its onset of action is 30-50 minutes and half life is 40 hours. The duration of action of drug is 24 hours. All undergo ...
Amlodipine is contraindicated in patient with known sensitivity to dihydropyridines or any component of the medication.
Use with caution in patients with hepatic cirrhosis. It should not be administered during pregnancy or lactation or to women of childbearing potential unless effective contraception is used. It is not recommended for children.
Headache, flushing, dizziness, palpitation and hypotension may occur during the first few hours after dosing as the plasma concentration is increasing, particularly if the initial dose is too high or increased too rapidly. The main side effects are odema and flushing and occur as a result of vasodil...
In clinical trials, amlodipine has been safely administered with 13-adrenergic blocking agents. Unlike other dihydro pyridine calcium-channel blockers, amlodipine showed no changes in pharmacokinetics when administered with grapefruit juice. Unlike other drugs metabolized by cytochrome P450, amlodi...
